Professor; Neurology; Boston University Medical Campus 715 Albany St, 560 Boston, Ma 02118 Timing: Fiscal Year 2002; Project Start 30-SEP-1997; Project End 31-JUL-2007 Summary: (provided by applicant): The cause of idiopathic Parkinson's disease (PD), a debilitating disease that afflicts an estimated 1 percent of persons age 60 or older, remains unknown. In this application, we propose genetic epidemiological studies, genetic linkage studies, and candidate gene studies for PD. In the past four years, we have established a ulti-institutional research program that has collected 300 PD affected sibling pairs and extended families.
However, their use has been limited by high doses of vectors needed, vector-cytotoxicity, short-term transgene expression, and deleterious effects of anti-adenovirus inflammatory and immune responses. These limitations are due to: (i) low efficiency of transcriptional promoter elements currently used, which directly leads to the need to use higher doses of vectors, and (ii) cytotoxicity and immunogenicity of viral proteins expressed from the genomes of first generation adenoviral vectors. We now wish to initiate the next stage in the design of efficient, safe, long-term expressing vectors for neurological gene therapy.
Studies with "F-fluoro-deoxyglucose (FDG) and PET have demonstrated an abnormal metabolic network in clinically non-manifesting carriers of Idiopathic Torsion Dystonia (ITD) DYT I gene. In preliminary studies, we have found that motor sequence learning may be impaired in these same gene carriers. We plan to complete the psychophysical studies in non-manifesting DYTI carriers and to assess the effect of GPiDBS on rcbf in affected DYTI patients. Specifically, we will ask the following questions: Will non-manifesting DYTI carriers show motor learning and network activation abnormalities that parallel their resting metabolic abnormalities?
